Bespoke promoters for safer and more effective cell and gene therapies
The right regulatory sequence can support cell type-specific, condition-responsive or dose-aware expression. The wrong one can limit safety, efficacy or durability before a therapy reaches its full potential.
SuRE™ is Annogen’s large-scale screening platform for promoter and enhancer development. It combines broad regulatory sequence exploration with sensitive experimental readouts in relevant biological systems, helping teams move from promoter assumptions to validated candidates.
You define the promoter profile your therapy needs: target cell type, expression strength, condition response and off-target constraints.
Based on the right in vitro or in vivo models, SuRE™ tests up to 300 million promoter and enhancer candidates in parallel, using proprietary and public sequence data where relevant. Subsequently, we combine the most promising elements and screen again.
We identify regulatory sequences that match the required expression behavior and help prioritize candidates for further validation.
You receive a set of top candidate elements, typically around ten validated sequences, that you can take forward and protect as your own IP. The exact deliverable is agreed with you at the start.
A bespoke promoter project typically takes 6-9 months to complete.
Test thousands of promoter and enhancer candidates in one experiment, instead of iterating through a few designs at a time.
Measure subtle differences in regulatory activity with the sensitivity needed to separate real signal from screening noise.
Screen in the systems that best reflect how your final constructs will be used, including primary cells, lentiviral configurations and AAV or episomal contexts.
Develop proprietary promoters around your therapeutic gene, target cell and required expression behavior.
Start from validated regulatory sequences when your therapy needs a faster route to expression control.
Identify regulatory DNA positions that can modulate endogenous gene expression toward therapeutic effect.
Move promoter screening closer to therapeutic reality.
SuRE™ can be used in lentiviral and AAV-based configurations, enabling parallel assessment of regulatory sequences in advanced therapy-relevant models including mouse, pig and NHP. This reveals promoter behavior that may not be visible in standard cell based models, or where in vitro models are not available.

Map genomic integration sites when transgene expression depends on genomic context as well as regulatory sequence design.
Annogen designs and optimises the gene regulatory elements, mainly promoters, that determine where, when and how strongly your gene of interest is expressed. Our SuRE platform measures the activity of millions of regulatory sequences directly in relevant cells or organisms, so the elements we deliver are validated by experiment rather than predicted. You decide the expression profile you need; we find and build the element that achieves it.
Hundreds of millions in a single experiment. That scale is the point: rather than testing a handful of candidate promoters, we survey the full design space and let the data show which elements perform best for your system.
The standard readout is transcriptional, measured by sequencing expressed barcodes, which lets us quantify very large libraries at once. Where a project needs it, we can discuss protein-level validation as a follow-up step.
We can design inducible systems around several independent triggers, and a TET-operator can be built in where you want it. If you are looking to avoid specific third-party components, that is worth raising early so we design accordingly.
Durability matters, so it is part of how we design rather than an afterthought. Expression stability depends on the element, the integration context and the cell type, and we can build longer-term readouts into a project where it is critical to your application.
Annogen's SuRE platform is species-agnostic. We have worked in human, non-human primate, mouse and various livestock, as well as crops such as corn, soy and tomato.
Both non-exclusive licences to existing elements and exclusive arrangements for bespoke work are possible. Which applies depends on the element and the scope, and we are glad to discuss the options directly.
